Item Open Access Application of the Ugi reaction with multiple amino acid-derived components: Synthesis and conformational evaluation of piperazine-based minimalist peptidomimetics(Royal Society of Chemistry, 2015) Stucchi, M.; Cairati, S.; Cetin-Atalay, R.; Christodoulou, M.S.; Grazioso G.; Pescitelli G.; Silvani, A.; Yildirim, D.C.; Lesma G.The concurrent employment of α-amino acid-derived chiral components such as aldehydes and α-isocyanoacetates, in a sequential Ugi reaction/cyclization two-step strategy, opens the door to the synthesis of three structurally distinct piperazine-based scaffolds, characterized by the presence of l-Ala and/or l-Phe-derived side chains and bearing appropriate functionalities to be easily applied in peptide chemistry. By means of computational studies, these scaffolds have been demonstrated to act as minimalist peptidomimetics, able to mimic a well defined range of peptide secondary structures and therefore potentially useful for the synthesis of small-molecule PPI modulators. Preliminary biological evaluation of two different resistant hepatocellular carcinoma cellular lines, for which differentiation versus resistance ability seem to be strongly correlated with well defined types of PPIs, has revealed a promising antiproliferative activity for selected compounds. © The Royal Society of Chemistry 2015.Item Open Access Electrochemical synthesis of mesoporous gold films toward mesospace-stimulated optical properties(Nature Publishing Group, 2015) Li C.; Dag Ö.; Dao, T.D.; Nagao, T.; Sakamoto, Y.; Kimura, T.; Terasaki O.; Yamauchi, Y.Mesoporous gold (Au) films with tunable pores are expected to provide fascinating optical properties stimulated by the mesospaces, but they have not been realized yet because of the difficulty of controlling the Au crystal growth. Here, we report a reliable soft-templating method to fabricate mesoporous Au films using stable micelles of diblock copolymers, with electrochemical deposition advantageous for precise control of Au crystal growth. Strong field enhancement takes place around the center of the uniform mesopores as well as on the walls between the pores, leading to the enhanced light scattering as well as surface-enhanced Raman scattering (SERS), which is understandable, for example, from Babinet principles applied for the reverse system of nanoparticle ensembles. © 2015 Macmillan Publishers Limited. The method is based on adsorption of Pd(II) ions, formation of a Pd catalyst, and autocatalytic deposition of the alloy from dissolved metal salts, reduced by a borane compound. Extensive energy-filtering TEM investigations at the nanoscale revealed that the synthesized wires are alloys of Co, Fe, and Ni. We confirmed by high-resolution TEM that our alloy nanowires are at least partially crystalline, which is compatible with typical Co-rich alloys. Ni traces bestow higher stability, presumably against corrosion, as also known from bulk CoFe. Alloy nanowires, as small as the ones presented here, might be used for a variety of applications including high density data storage, imaging, sensing, and even drug delivery. © 2012 IOP Publishing Ltd.Item Open Access Environmental friendly InP/ZnS nanocrystals(2012) Coşkun, YaseminSemiconductor nanocrystals are nanometer scale fluorescent crystallites with tunable optical properties, which can be controlled by the material composition and particle size. They can be prepared using various synthesis techniques and find applications in many different areas ranging from life sciences to electronics. In this thesis, indium phosphide based nanocrystals are studied for LED applications. The thesis research work focuses on the colloidal synthesis method and material characterization of these nanocrystals. Using one pot synthesis method, the indium phosphide/zinc sulfide (InP/ZnS) core/shell nanocrystal structures are synthesized. This synthesis technique allows for a reproducible and tunable preparation method. The material characterization techniques used in this thesis include UV-Vis spectroscopy, photoluminescence spectroscopy, transmission electron microscopy (TEM), X-ray diffraction, X-ray photoelectron spectroscopy (XPS), inductively coupled plasma-mass spectrometry (ICP-MS), and Zeta Sizer (or DLS). These properties make InP/ZnS nanocrystals comparable to their cadmium (Cd) containing counterparts with respect to their optical properties, and InP/ZnS nanocrystals offer the potential to replace them because of environmental concerns in LED applications.Item Open Access In situ synthesis of biomolecule encapsulated gold-cross-linked poly(ethylene glycol) nanocomposite as biosensing platform: A model study(Elsevier BV, 2010) Odaci, D.; Kahveci, M.U.; Sahkulubey, E.L.; Ozdemir, C.; Uyar, Tamer; Timur, S.; Yagci Y.In situ synthesis of poly(ethylene glycol) (PEG) hydrogels containing gold nanoparticles(AuNPs) and glucose oxidase (GOx) enzyme by photo-induced electron transfer process was reported here and applied in electrochemical glucose biosensing as the model system. Newly designed bionanocomposite matrix by simple one-step fabrication offered a good contact between the active site of the enzyme and AuNPs inside the network that caused the promotion in the electron transfer properties that was evidenced by cyclic voltammetryas well as higher amperometric biosensing responses in comparing with response signals obtained from the matrix without AuNPs. As well as some parameters important in the optimization studies such as optimum pH, enzyme loading and AuNP amount, the analytical characteristics of the biosensor (AuNP/GOx) were examined by the monitoring of chronoamperometric response due to the oxygen consumption through the enzymatic reaction at − 0.7 V under optimized conditions at sodium acetate buffer (50 mM, pH 4.0) and the linear graph was obtained in the range of 0.1–1.0 mM glucose. The detection limit (LOD) of the biosensor was calculated as 0.06 mM by using the signal to noise ratio of 3. Moreover, the presence of AuNPs was visualized by TEM. Finally, the biosensor was applied for glucose analysis for some beverages and obtained data were compared with HPLC as the reference method to test the possible matrix effect due to the nature of the samples.Item Open Access Synthesis and anticancer activity evaluation of some benzothiazole-piperazine derivatives(Bentham Science Publishers B.V., 2015) Gurdal, E.E.; Buclulgan, E.; Durmaz I.; Cetin-Atalay, R.; Yarim, M.Synthesis, characterization and cytotoxic activities of ten benzothiazole-piperazine derivatives were reported. In vitro cytotoxic activities of compounds were screened against hepatocellular (HUH-7), breast (MCF-7) and colorectal (HCT-116) cancer cell lines by sulphorhodamine B assay. Based on the GI50 values of the compounds, most of the benzothiazole-piperazine derivatives are active against HUH-7, MCF-7 and HCT-116 cancer cell lines. Aroyl substituted compounds 1h and 1j were found to be the most active derivatives. In addition, further investigation of compounds 1h and 1j by Hoechst staining and FACS revealed that these compounds cause apoptosis by cell cycle arrest at subG1 phase. © 2015 Bentham Science Publishers.Item Open Access Synthesis and dye sensitized solar cell applications of Bodipy derivatives with bis-dimethylfluorenyl amine donor groups(Royal Society of Chemistry, 2015) Çakmak, Y.; Kolemen, S.; Buyuktemiz, M.; Dede, Y.; Erten-Ela, S.Three Bodipy dyes with strong absorptivities in the visible and near infrared regions were designed, synthesized and their potential as photosensitizers for liquid electrolyte-based dye sensitized solar cells have been evaluated. For the first time Bodipy derivatives with bis-dimethylfluorenyl amine donor groups which were known for their bulky structures as donor groups have been used together. We altered our mostly used triphenylamine group with these and investigated the dye-sensitized solar cell efficiencies of this new class of Bodipy dyes. © The Royal Society of Chemistry and the Centre National de la Recherche Scientifique 2015.Item Open Access Synthesis of novel substituted purine derivatives and identification of the cell death mechanism(Elsevier Masson SAS, 2015) Demir, Z.; Guven, E.B.; Ozbey, S.; Kazak, C.; Atalay, R.C.; Tuncbilek, M.Novel substituted adenine and purine derivatives were designed and synthesized.Compound 36 displayed the greatest cytotoxic activity with IC50 less than 1 1/4M.36 induces senescence associated cell death, which was demonstrated with SA2-Gal assay. © 2014 Elsevier Masson SAS.
