Item Open Access Chitosan scaffolds with BMP-6 loaded alginate microspheres for periodontal tissue engineering(2012) Soran, Z.; Aydin, R.S.T.; Gumusderelioglu, M.The aim of this study is to develop an effective growth factor releasing scaffold-microsphere system for promoting periodontal tissue engineering. Bone morphogenetic protein-6 (BMP-6)-loaded alginate microspheres in narrow size distribution were produced by optimising electrospraying conditions. The addition of these microspheres to chitosan gels produced a novel scaffold in which not only the pore sizes and interconnectivity were preserved, but also a controlled release vehicle was generated. Loading capacity was adjusted as 50ng or 100ng BMP-6 for each scaffold and the controlled release behaviour of BMP-6 from chitosan scaffolds was observed during seven days. Cell culture studies were carried out with rat mesenchymal stem cells derived from bone marrow in three groups; chitosan scaffolds, chitosan scaffolds containing BMP-6-loaded alginate microspheres and chitosan scaffolds with free BMP-6 in culture medium. Results showed that controlled delivery of BMP-6 from alginate microspheres has a significant effect on osteogenic differentiation. © 2012 Informa UK Ltd All rights reserved.Item Open Access Microsphere-Assisted Super-Resolved Mirau Digital Holographic Microscopy for Cell Identification(OSA - The Optical Society, 2017) Aakhte, M.; Abbasian, V.; Akhlaghi, E. R.; Anand, A.; Javidi, B.In this paper, we use a glass microsphere incorporated into a digital holographic microscope to increase the effective resolution of the system, aiming at precise cell identification. A Mirau interferometric objective is employed in the experiments, which can be used for a common-path digital holographic microscopy (DHMicroscopy) arrangement. High-magnification Mirau objectives are expensive and suffer from low working distances, yet the commonly used low-magnification Mirau objectives do not have high lateral resolutions. We show that by placing a glass microsphere within the working distance of a low-magnification Mirau objective, its effective numerical aperture can be increased, leading to super-resolved three-dimensional images. The improvement in the lateral resolution depends on the size and vertical position of microsphere, and by varying these parameters, the lateral resolution and magnification may be adjusted. We used the information from the super-resolution DHMicroscopy to identify thalassemia minor red blood cells (tRBCs). Identification is done by comparing the volumetric measurements with those of healthy RBCs. Our results show that microsphere-assisted super-resolved Mirau DHMicroscopy, being common path and off-axis in nature, has the potential to serve as a benchtop device for cell identification and biomedical measurements.Item Open Access Newly designed silver coated-magnetic, monodisperse polymeric microbeads as SERS substrate for low-level detection of amoxicillin(Elsevier, 2016-09) Kibar, G.; Topal, A. E.; Dana, A.; Tuncel, A.We report the preparation of silver-coated magnetic polymethacrylate core-shell nanoparticles for use in surface-enhanced Raman scattering based drug detection. Monodisperse porous poly (mono-2-(methacryloyloxy)ethyl succinate-co-glycerol dimethacrylate), poly (MMES-co-GDMA) microbeads of ca. 5 μm diameter were first synthesized through a multistage microsuspension polymerization technique to serve as a carboxyl-bearing core region. Microspheres were subsequently magnetized by the co-precipitation of ferric ions, aminated through the surface hydroxyl groups and decorated with Au nanoparticles via electrostatic attraction. An Ag shell was then formed on top of the Au layer through a seed-mediated growth process, resulting in micron-sized monodisperse microbeads that exhibit Raman enhancement effects due to the roughness of the Ag surface layer. The core-shell microspheres were used as a new substrate for the detection of amoxicillin at trace concentrations up to 10-8 M by SERS. The proposed SERS platform can be evaluated as a useful tool for the follow-up amoxicillin pollution and low-level detection of amoxicillin in aqueous media.
