Item Open Access Baudelaire'in çift görünümlü oda'sında yansımalar(Ürün Yayınları, 2004) Sunel, A. Hamitin this research the author translated "La Chambre Double" the fıfth poem in Baudelaire's book of "Petits Poemes en Prose". in this research, the author tries to analyze the poem on the basis of the original poet's system of thought by establishing similarities between the descriptions of the room and some events in life. in both descriptions of the room, the author tries to explain that every word and every sentence reflects a universal point of view of the original poet and that every event correspond with something in the poet's real life. The author, at the same time, wants to know who the idol that foverned imaginary throne of pleasure was. He also wishes to investigate who the spectre and the disguised persons are. Moreover, he attempts to explain why the poet considers the bottle of laudanum an old and dreadful friend and to which events, the description of the room refers.Item Open Access Glycosylation: a tool for engineering protein based materials(2019-09) Işılak, Musa EfeBiofilms are bacterial communities formed by adhesion of cells to each other via extracellular matrix. Proteins, one of the components of biofilm matrix, can form fibers and have properties enabling their use as materials. Bacillus subtilis biofilm major protein TasA is a fiber forming non-amyloidogenic protein providing biofilm rigidity. Glycosylation is a post-translational modification observed in all domains of life where sugar groups are added on proteins covalently. Campylobacter jejuni glycosylation is the first bacterial N-linked glycosylation discovered and the most studied system where a heptasaccharide is attached to protein. C. jejuni glycosylation has variety of functions among adhesion, protease resistance and thermal stability. Glycosylation may become an additional method for engineering biofilm proteins as materials with unique properties. For this purpose, we examined effect of glycosylation on structure of natively non-glycosylated proteins by glycosylating alkaline phosphatase enzyme (ALP) at different locations on the protein. Interestingly, phosphatase activity assay showed slight increase in ALP activity on pure proteins glycosylated at the C-terminus. Next, TasA protein was glycosylated at the C-terminus. Glycosylation had no significant effect on fibrillation of TasA in vitro. Secondary structure analysis using circular dichroism data revealed shift from antiparallel to helix structure with glycosylation. Quartz crystal microbalance experiments indicated increased adhesive properties on glycosylated TasA protein on gold. Its application possibility as cell adhesive was assessed by visualizing surface coverage of polystyrene cell culture plate under scanning electron microscope. As a consequence, glycosylation was used as an engineering method for protein-based material development for the first time.Item Open Access The relationship of screen material with visitor behavior in exhibition spaces(2022-04) Karacan, EvrimThe screen, with its diverse forms and materials, has been a part of the exhibition space as art practice as well as an integral part of human beings' lives. The purpose of this study is to investigate the impact of the screen materials on space formation through observing different screen materials and explore the relationship between the materials of screens and visitor, by addressing visitor behavior that is divided as attention and interaction in exhibition spaces. Previous studies were mostly elaborated in the field of media and art, and few research was found in the architecture field. Therefore, this study seeks to present a comprehensive approach by gathering media, art, and architecture perspectives together in the visitor studies in the context of exhibition spaces. An exhibition experiment was designed, and different instruments were used in the assessment of the participants’ behavior, experience, and feelings. A total of 32 participants were selected based on defined demographic parameters. The findings of this study presented that, the profession item showed a significant correlation with the screen preferences and screen materials have an impact on the architectural layout from visitors’ perspective in terms of its physical form and its influence on the layout. Lastly, screen materials have a significant impact on the amount of attention paid as well as on the number of visitor interactions with exhibit objects in exhibition spaces.Item Open Access Sterilization of PMMA microfluidic chips by various techniques and investigation of material characteristics(Elsevier, 2016) Yavuz, C.; Oliaei, S. N. B.; Cetin, B.; Yesil-Celiktas, O.The sterilization of microfluidic chips is a vital step of the fabrication process prior to the customer use in biomedical applications. The aim of this study was to analyze the influence of different sterilization techniques and to compare the characteristics of the material before and after sterilization of polymethylmethacrylate (PMMA) microchips. For this, supercritical carbon dioxide (SC-CO2) along with standard sterilization methods such as ultraviolet (UV), heat (autoclaving), ethylene oxide (EtO) and hydrogen peroxide (H2O2) were applied. The treated microchips were analyzed by Scanning Electron Microscopy, Differential Scanning Calorimetry, Fourier Transform Infrared Spectroscopy and Laser Scanning Microscopy in order to ascertain any changes in the chemical structure and surface morphology. The optimum sterilization parameters for SC-CO2 were elicited as 120 bar, 40°C and 60 min which provided complete sterility and did not alter the main properties of the polymer along with EtO and H2O2 sterilizations unlike heat and UV treatments. However, surface roughness and microchannel profiles were negatively affected. Although complete sterility was achieved, each protocol has its own strengths and weaknesses. © 2015 Elsevier B.V. Materials and colors rarely appear alone in interiors; therefore, in the scope of this study, the researchers explored material pairs and color pairs in addition to single materials and single colors. To elicit free associations from these interior design elements, 192 randomly selected volunteers participated in an experiment using a group of material (fabric, timber, plasterboard) and color (red, green, white) models under controlled conditions. The results contribute to an increased understanding of the associations between the concepts of materials and colors in interiors. While, each model was associated with sensory descriptors, only some models were associated with symbolic or affective descriptors. Single materials were related to different descriptors in interiors on their own, but when they were paired they were associated with fewer affective descriptors. The results showed that color pairs were always associated with all types of descriptors with an exception of red and green color pair, which was not mentioned with affective descriptors. The study findings are expected to be beneficial for interior architects, architects, product designers and researchers who want to shape and investigate a user's experience of interiors. © 2016 Wiley Periodicals, Inc. Warmth is a prominent characteristic of interior architecture and is related to colours and materials. Although the effects of single colours and single materials on warmth have been explored, colours and materials rarely appear alone in interiors and there has been no research on how paired colours and paired materials affect warmth perception in interiors. Therefore, the main aim of this study is to investigate their effects through a seven-point semantic differential scale and open ended questions.192 different participants assessed three different colours (red, white, and green), and their pairs or three different materials (fabric, timber, and plasterboard), and their pairs under controlled conditions. Findings demonstrated that single colours and paired colours both affect warmth perception in interiors. The effects of single colours in interiors are subtle in warmth perception: red is perceived warmer than green and green is perceived warmer than white. All single colours have a moderate level of warmth in interiors as pairs, consequently red (warm colour) appears to increase and white (achromatic colour) appears to decrease the warmth perception of their pairs. Furthermore, as single materials timber and fabric have the same level of warmth and are warmer than plasterboard whereas there is not any difference between pairs. Findings indicated that natural materials are perceived warmer than artificial one.
