Item Open Access Bioactive glycopeptide nanofibers for tissue regeneration applications(2016-05) Çalışkan, Özüm Şehnaz.Natural extracellular matrix (ECM) is rich in glycopeptides and glycosaminoglycans, which function in controlling cellular processes. In this thesis, glycopeptide molecules that mimic natural glycopeptides and glycosaminoglycans were designed and synthesized and it was demonstrated that they induce directed differentiation of mesenchymal stem cells into chondrogenic and adipogenic lineages. In the first part of the study, hyaluronic acid (HA)-mimicking glycopeptide amphiphile molecules were synthesized to induce chondrogenic differentiation of mesenchymal stem cells (MSC). HA is the most abundant glycosaminoglycan (GAG) found in hyaline cartilage ECM. Peptide amphiphiles were synthesized by solid phase peptide synthesis method and used to form self-assembled bioactive glycopeptide nanofibers which mimic fibrous morphology of the ECM. Scanning electron microscopy (SEM), transmission electron microscopy (TEM), and circular dichroism (CD) were used for morphology and secondary structure analyses of the obtained nanofibers. It was demonstrated that glycopeptide amphiphiles create fibrous structure formed by nanofibers. Morphological changes, GAG production (Safranin-O staining and DMMB analysis), and chondrogenic gene marker expressions (qRT-PCR) of MSCs cultured on HA-mimetic nanofibers were analyzed. It was shown that HA-mimetic glycopeptide nanofibers induce early differentiation of MSCs into hyaline like chondrocytes. In the second part of the study, it was demonstrated that minor changes on glycopeptide backbone can create specific glycopeptides which induce differentiation of MSCs into brown adipocytes. Brown fat adipocytes do not store chemical energy as fat but dissipates it as heat and so they have emerged as promising anti-obesity agents. Lipid droplet accumulation (Oil Red-O staining) and adipogenic gene marker expression analyses (qRT-PCR) showed that the new glycopeptide nanofiber scaffold is a specific inducer of differentiation of MSCs into brown fat adipocytes.Item Open Access Bioactive peptide functionalized aligned cyclodextrin nanofibers for neurite outgrowth(Royal Society of Chemistry, 2017) Hamsici, S.; Cinar, G.; Celebioglu A.; Uyar, Tamer; Tekinay, A. B.; Güler, Mustafa O.Guidance of neurite extension and establishment of neural connectivity hold great importance for neural tissue regeneration and neural conduit implants. Although bioactive-epitope functionalized synthetic or natural polymeric materials have been proposed for the induction of neural regeneration, chemical modifications of these materials for neural differentiation still remain a challenge due to the harsh conditions of chemical reactions, along with non-homogeneous surface modifications. In this study, a facile noncovalent functionalization method is proposed by exploiting host-guest interactions between an adamantane-conjugated laminin derived bioactive IKVAV epitope and electrospun cyclodextrin nanofibers (CDNFs) to fabricate implantable scaffolds for peripheral nerve regeneration. While electrospun CDNFs introduce a three-dimensional biocompatible microenvironment to promote cellular viability and adhesion, the bioactive epitopes presented on the surface of electrospun CDNFs guide the cellular differentiation of PC-12 cells. In addition to materials synthesis and smart functionalization, physical alignment of the electrospun nanofibers guides the cells for enhanced differentiation. Cells cultured on aligned and IKVAV functionalized electrospun CDNFs had significantly higher expression of neuron-specific βIII-tubulin and synaptophysin. The neurite extension is also higher on the bioactive aligned scaffolds compared to random and non-functionalized electrospun CDNFs. Both chemical and physical cues were utilized for an effective neuronal differentiation process. © The Royal Society of Chemistry.Item Open Access Enhanced angiogenic effects of RGD, GHK peptides and copper (II) compositions in synthetic cryogel ECM model(Elsevier, 2020) Zoughaib, M.; Luong, D.; Garifullin, Ruslan; Gatina, D. I.Synthetic oligopeptides are a promising alternative to natural full-length growth factors and extracellular matrix (ECM) proteins in tissue regeneration and therapeutic angiogenesis applications. In this work, angiogenic properties of dual and triple compositions containing RGD, GHK peptides and copper (II) ions (Cu2+) were for the first time studied. To reveal specific in vitro effects of these compositions in three-dimensional scaffold, adamantyl group bearing peptides, namely Ada-Ahx-GGRGD (1) and Ada-Ahx-GGGHK (2), were effectively immobilized in bioinert pHEMA macroporous cryogel via host-guest β-cyclodextrin-adamantane interaction. The cryogels were additionally functionalized with Cu2+ via the formation of GHK-Cu complex. Angiogenic responses of HUVECs grown within the cryogel ECM model were analyzed. The results demonstrate that the combination of RGD with GHK and further with Cu2+ dramatically increases cell proliferation, differentiation, and production of a series of angiogenesis related cytokines and growth factors. Furthermore, the level of glutathione, a key cellular antioxidant and redox regulator, was altered in relation to the angiogenic effects. These results are of particular interest for establishing the role of multiple peptide signals on regeneration related processes and for developing improved tissue engineering materials.Item Open Access A glycosaminoglycan mimetic peptide nanofiber gel as an osteoinductive scaffold(Royal Society of Chemistry, 2016) Tansik, G.; Kilic, E.; Beter, M.; Demiralp, B.; K.Sendur, G.; Can, N.; Ozkan, H.; Ergul, E.; Güler, Mustafa O.; Tekinay, A. B.Biomineralization of the extracellular matrix (ECM) plays a crucial role in bone formation. Functional and structural biomimetic native bone ECM components can therefore be used to change the fate of stem cells and induce bone regeneration and mineralization. Glycosaminoglycan (GAG) mimetic peptide nanofibers can interact with several growth factors. These nanostructures are capable of enhancing the osteogenic activity and mineral deposition of osteoblastic cells, which is indicative of their potential application in bone tissue regeneration. In this study, we investigated the potential of GAG-mimetic peptide nanofibers to promote the osteogenic differentiation of rat mesenchymal stem cells (rMSCs) in vitro and enhance the bone regeneration and biomineralization process in vivo in a rabbit tibial bone defect model. Alkaline phosphatase (ALP) activity and Alizarin red staining results suggested that osteogenic differentiation is enhanced when rMSCs are cultured on GAG-mimetic peptide nanofibers. Moreover, osteogenic marker genes were shown to be upregulated in the presence of the peptide nanofiber system. Histological and micro-computed tomography (Micro-CT) observations of regenerated bone defects in rabbit tibia bone also suggested that the injection of a GAG-mimetic nanofiber gel supports cortical bone deposition by enhancing the secretion of an inorganic mineral matrix. The volume of the repaired cortical bone was higher in GAG-PA gel injected animals. The overall results indicate that GAG-mimetic peptide nanofibers can be utilized effectively as a new bioactive platform for bone regeneration. © 2016 The Royal Society of Chemistry.Item Open Access Heparin mimetic peptide nanofiber gel promotes regeneration of full thickness burn injury(Elsevier Ltd, 2017) Yergoz, F.; Hastar, N.; Cimenci, C. B.; Tekinay, T.; Güler, Mustafa O.Burn injuries are one of the most common types of trauma worldwide, and their unique physiology requires the development of specialized therapeutic materials for their treatment. Here, we report the use of synthetic, functional and biodegradable peptide nanofiber gels for the improved healing of burn wounds to alleviate the progressive loss of tissue function at the post-burn wound site. These bioactive nanofiber gels form scaffolds that recapitulate the structure and function of the native extracellular matrix through signaling peptide epitopes, which can trigger angiogenesis through their affinity to basic growth factors. In this study, the angiogenesis-promoting properties of the bioactive scaffolds were utilized for the treatment of a thermal burn model. Following the excision of necrotic tissue, bioactive gels and control solutions were applied topically onto the wound area. The wound healing process was evaluated at 7, 14 and 21 days following injury through histological observations, immunostaining and marker RNA/protein analysis. Bioactive peptide nanofiber-treated burn wounds formed well-organized and collagen-rich granulation tissue layers, produced a greater density of newly formed blood vessels, and exhibited increased re-epithelialization and skin appendage development with minimal crust formation, while non-bioactive peptide nanofibers and the commercial wound dressing 3M™ Tegaderm™ did not exhibit significant efficiency over sucrose controls. Overall, the heparin-mimetic peptide nanofiber gels increased the rate of repair of burn injuries and can be used as an effective means of facilitating wound healing.Item Open Access Materials for articular cartilage regeneration(Bentham Science Publishers B.V., 2012) Tombuloglu, Ayşegül; Tekinay, Ayşe B.; Güler, Mustafa O.Many health problems remaining to be untreatable throughout the human history can be overcome by utilizing new biomedical materials. Healing cartilage defects is one of the problems causing significant health issue due to low regeneration capacity of the cartilage tissue. Scaffolds as three-dimensional functional networks provide promising tools for complete regeneration of the cartilage tissue. Diversity of materials and fabrication methods give rise to many forms of scaffolds including injectable and mechanically stable ones. Various approaches can be considered depending on the condition of cartilage defect. A scaffold should maintain tissue function within a short time, and should be easily applied in order to minimally harm the body. This review will cover several patents and other publications on materials for cartilage regeneration with an outlook on essential characteristics of materials and scaffolds.Item Open Access Nanosized delivery systems for tissue regeneration(John Wiley & Sons, 2016-03-11) Çınar, Göksu; Mumcuoğlu, Didem; Tekinay, Ayşe B.; Güler, Mustafa O.; Güler, Mustafa O.; Tekinay, Ayşe B.This chapter focuses on advanced delivery of biologics including growth factors (GFs), cytokines, genes, or siRNAs using a variety of nanosized systems for different regeneration applications focusing on bone, cartilage, nervous system, and muscle regeneration strategies. Gene therapy provides permanent genetic alteration, but only transient actions of protein therapeutics can be needed for tissue regeneration applications. The limitations of biologics delivery and alternative strategies for overcoming recent problems are underlined presenting recent examples from the literature. In addition, specific targeting and cellular internalization strategies of biologies delivery for tissue regeneration are discussed for providing future perspectives to the readers in this field. Overall, it is believed that advanced nanosized delivery systems integrated with multicomponent designs will open new opportunities in delivery technologies and strategies for tissue regeneration.
